How do I check transmission fluid on a 2000 sportster 1200

The transmission and the primary case use the same reservoir. There should be instructions in the owners manual or better yet get the factory service manual.
On my 85 Ironhead there is a hole at the level at which the fluid should be maintained. With the bike sitting level You remove the screw from that hole and oil should barely trickle out. When refilling you remove the primary chain inspection plug and pour in oil until it runs out the level hole and stops. I fudge a little and put the screw in before it stops so that she is just slightly overfilled, gives just a little reserve. I think your bike has a derby cover so they might have you check it there, that's why a manual is advisable.
Make sure you use the proper oil, I've always used Harley Sporttrans, I think they may have renamed it, ask your parts man to make sure.

It has been re-named, now it's called Formula Plus, or something close to that. As Mike just stated, if your bike has the inspection cover, you check it with the cover off. Also, if your bike has the primary chain inspection cover, you can fill thru there if you don't want to pull the clutch inspection cover off.( at least you can on mine, an 05 xl1200). I just pour the full bottle of fluid in the hole, all 32 oz and button it back up!

In addition, proper level on your 2000 is to the bottom of the derby cover opening with the bike vertical, the manual calls for a full quart, however on mine I've learned to short it by one ounce, if I don't it puffs it out the transmission breather tube across the side of the fender. Good luck.
